INTENDED USE
- TraceMedia™ TCBS Agar is a pre-poured petri dish medium of 90mm diameter used for the isolation and selection of Vibrio species. It allows the differentiation of Vibrio cholerae from other Vibrio species based on their ability to ferment sucrose.
PRINCIPLE
- TraceMedia™ TCBS Agar is designed for the isolation and selection of Vibrio cholerae and pathogenic Vibrio species associated with intestinal diseases. Thiosulfate, sodium citrate, and the alkaline pH of the medium significantly inhibit the growth of Enterobacteria.
- Beef extract and sodium cholate slow down the growth of enterococci and inhibit the growth of gram-positive bacteria.
- The medium’s acidic pH is the result of the sucrose fermentation by Vibrio, causing the bromothymol blue indicator to turn yellow.
- Bromothymol blue and thymol blue act as pH indicators. The presence of ferric citrate detects the production of hydrogen sulfide from thiosulfate as a sulfur source.
- Peptone and yeast extract provide nitrogen, vitamins, and amino acids in TCBS agar.
- Sodium chloride supports optimal growth and metabolic activity of Vibrio species living in a saline environment.
- Agar is used as a solidifying agent. The increased pH is used to enhance the growth of Vibrio cholerae, as this organism is sensitive to acidic conditions.
INTERPRETATION
- After incubation for the required time, usually 18-24 hours, observe the colonies developing on the surface of the agar plate.
- Further identification of the isolated bacteria must be conducted through appropriate tests.
