INTENDED USE
- BacterTube™ Nitrate Broth is used to detect the ability of bacteria to reduce nitrate.
PRINCIPLES
- BacterTube™ Nitrate Broth is a basic medium that includes potassium nitrate, which is reduced by the nitroreductase enzyme of microorganisms. Some microorganisms reduce nitrate to nitrite, while others reduce nitrate to NH₃, N₂, etc. The reduction of nitrate to nitrite is detected by adding sulfanilic acid (reagent A) and N,N-dimethyl-1-naphthylamine (reagent B). Sulfanilic acid reacts with nitrite to form a diazonium salt, which then combines with N,N-dimethyl-1-naphthylamine to form a red-colored complex. Other forms of nitrate reduction products (NH₃, N₂, etc.) may not show the red color change, which can be confirmed by adding zinc powder (reagent C).
