BacterTube™ Nitrate Broth

INTENDED USE

BacterTube™ Nitrate Broth is used to detect the ability of bacteria to reduce nitrate.

PRINCIPLES

BacterTube™ Nitrate Broth is a basic medium that includes potassium nitrate, which is reduced by the nitroreductase enzyme of microorganisms. Some microorganisms reduce nitrate to nitrite, while others reduce nitrate to NH₃, N₂, etc. The reduction of nitrate to nitrite is detected by adding sulfanilic acid (reagent A) and N,N-dimethyl-1-naphthylamine (reagent B). Sulfanilic acid reacts with nitrite to form a diazonium salt, which then combines with N,N-dimethyl-1-naphthylamine to form a red-colored complex. Other forms of nitrate reduction products (NH₃, N₂, etc.) may not show the red color change, which can be confirmed by adding zinc powder (reagent C).

Prior to inoculation, allow medium to equilibrate to room temperature
Use a sterile inoculation loop to transfer a sample from an isolated colony on the agar plate into the test tube containing the medium.
Incubate the tube at 35°C, Aerobic for 18 – 48 hours.
Examine the tube after 18 – 24 hours and again after 42 – 48 hours of incubation and gas production in the Durham tube.
Observe and record the results.

RESULTS

Positive nitrate reduction: A red color appears after adding reagents A and B to the test tube, or no color change occurs after adding zinc powder.
Negative nitrate reduction: No color change occurs after adding reagents A and B, and a red color appears after adding zinc powder.
