BacterTube™ Tryptic Soy Broth (TSB) 15% Glycerol is a liquid medium used in the long-term frozen storage of microorganisms.

PRINCIPLES

BacterTube™ Tryptic Soy Broth (TSB) 15% Glycerol is a highly nutritious medium used for cultivation of a wide variety of organisms. The combination of Tryptone and soya peptone makes the medium nutritious by providing nitrogenous, carbonaceous substances, amino acids and long chain peptides for the growth of microorganisms. Dextrose/glucose serve as the carbohydrate source and dibasic potassium phosphate buffer the medium. Sodium chloride maintains the osmotic balance of the medium. Glycerol plays a major role as an antifreezing agent. Hence it is recommended for maintenance of cultures as glycerol serves as an cryopreservant.

Prepare an overnight culture of the organism to be stored on an appropriate non-selective solid medium.
Using a sterile swab or inoculating loop, remove fresh growth from the culture medium and suspend the colonies in the tube to achieve the desired concentration of viable cells.
Shake or vortex well before freezing suspension at −20°C or below.

RESULTS

Appearance: The medium is homogeneous in color, free from particles.
Color: Light yellow, clear.
Sterility: No bacterial growth observed after incubation at 35–37°C for 48–72 hours.
