BacterTube™ Urea broth

INTENDED USE

BacterTube™ Urea Broth is recommended for the identification of bacteria based on urea utilization, particularly for differentiating Proteus, Salmonella, and Shigella species from clinical and non-clinical specimens.

PRINCIPLES

This medium differentiates bacteria based on their ability to hydrolyze urea using the enzyme urease:
- Urea hydrolysis releases ammonia, increasing the medium’s pH, which changes the indicator color to pink/red.
- The high buffering capacity and minimal nutrients prevent false positives caused by protein hydrolysis.
- A negative control without urea ensures accurate results by ruling out false positive

Prior to inoculation, allow medium to equilibrate to room temperature
Use a sterile inoculation loop to transfer a sample from an isolated colony on the agar plate into the test tube containing the medium.
Incubate the tube at 35°C.
Examine the tube after 6 – 8 hours and again after 12 – 18 hours of incubation.
