INTENDED USE
  • TraceMedia™ Chocolate Agar + Bacitracin is a prepared 90mm agar plate used for selective cultivation of Haemophilus influenzae. It is employed for culturing respiratory specimens.

PRINCIPLE

  • Special peptone provides essential nutrients, and sodium chloride acts as a pH buffer while starch neutralizes toxic fatty acids that may be present in the agar.
  • Isovitalex is a supplement providing factor V (nicotinamide adenine dinucleotide, NAD) for Haemophilus species.
  • Bacitracin is a polypeptide antibiotic that inhibits gram-positive bacteria, Neisseria, and Haemophilus spp.
READING THE RESULTS
  • After incubation for the required time, observe the growth of colonies on the surface of the agar plate and their characteristics.
  • Further identification of isolated bacteria must be conducted through appropriate tests.
  1. Samples related will be directly inoculated onto the agar plate.
  2. If the agar plate has been refrigerated, allow it to warm up to room temperature before inoculating the samples.
  3. Inoculate the samples onto the agar plate.
  4. Incubate the samples at 35 – 37°C, 5% CO2 for 16 – 24 hours.
