INTENDED USE
  • TraceMedia™ Sheep Blood Agar is a medium used for cultivation and isolation of various microorganisms.
  • It is also used to differentiate bacterial hemolysis types: alpha, beta, and gamma.
  • The medium is employed for culturing and isolating infectious agents from respiratory specimens, throat swabs, nasal secretions, etc.

PRINCIPLE

  • TraceMedia™ Sheep Blood Agar consists of Columbia Blood Agar Base supplemented with sheep blood, creating a medium that provides growth factors and allows for the determination of bacterial hemolytic reactions.
  • On the surface of the agar plate, bacteria with a clear zone of hemolysis are classified as beta hemolysis.
  • Bacteria that partially degrade hemoglobin, creating a greenish zone around the colonies, are classified as alpha hemolysis.
  • Gamma hemolysis refers to no hemolysis observed on the surface.
READING THE RESULTS
  • After incubation for the required time, observe the growth of colonies on the agar plate and their characteristics.
  • Further identification of isolated bacteria must be conducted through appropriate tests.
  1. Samples related will be directly inoculated onto the agar plate.
  2. If the agar plate has been refrigerated, allow it to warm up to room temperature before inoculating the samples.
  3. Inoculate the samples onto the agar plate.
  4. Incubate the samples at 35 – 37°C, 5% CO2 for 16 – 24 hours.
