INTENDED USE
- TraceMedia™ Hektoen Enteric Agar is a pre-poured agar medium in a 90mm petri dish used for selection and differentiation of pathogenic enteric bacteria from gram-negative enteric bacterial specimens.
PRINCIPLE
- TraceMedia™ Hektoen Enteric Agar contains a high peptone content to reduce the inhibitory effect of bile salts on Shigella strains.
- Additional carbohydrates (sucrose and salicin) provide better differentiation than lactose alone, and the low toxicity of the two indicator agents (acid fuchsin, bromothymol blue) enhances bacterial colony formation.
- Increased lactose content helps early detection of bacteria with slow lactose fermentation. Thiosulphate and Ferric citrate are included to detect bacteria that produce H2S.
RESULT
- After incubation for the required time, typically 18-24 hours, bacterial colonies are observed on the surface of the agar plate.
- Further identification of isolated bacteria must be conducted using appropriate tests.
The following plates are directly streaked:
- If the agar plates have been refrigerated, allow them to warm up to room temperature before streaking.
- Inoculate the samples onto the agar plates.
- Incubate at aerobic conditions at a temperature of 35 – 37°C for 16 – 24 hours.
