INTENDED USE
- BacterTube™ Selectine Cysteine is recommended as a selective enrichment medium for isolating Salmonella and possibly Shigella sonnei from samples such as feces, urine, water, and food.
PRINCIPLES
- The medium contains selenite, which inhibits the growth of competing bacteria while allowing the selective enrichment of Salmonella.
- Tryptone provides essential nutrients, including nitrogen and vitamins.
- Lactose helps maintain pH stability.
- L-Cystine enhances the recovery of Salmonella.
- After enrichment, the broth is subcultured on differential agar media for further isolation
- BacterTube™ Selectine Cysteine can be used with the following specimens: faeces, urine, water samples and foods
- Allow tubes to reach room temperature before inoculation
- Add 1g of stool sample or 1ml of stool suspension into the tube. The swab sample can be directly placed into the tube containing the medium.
- Completely dissolve the mixture in the tube.
- Incubate under aerobic conditions for 12 – 24 hours at 35 – 37°C.
- After incubation, transfer 1 – 2 drops from the tube onto selective media such as MacConkey Agar or Hektoen Enteric Agar to isolate colonies.
- Continue incubating at 35–37°C.
- Escherichia coli and Proteus species are not inhibited indefinitely. If the initial concentration of these microorganisms is high, it is best to subculture onto solid media after 6 hours of incubation in selenite broth.
