INTENDED USE

BacterBag™ Dry Half Fraser Broth is a high-quality dry medium, packaged in a transparent, lightweight, and convenient plastic bag, designed to simplify the preparation steps in microbiological culturing processes. Each BacterBag™ Dry Half Fraser Broth, when supplemented with sterile distilled water, produces 20L of complete medium, ready to use without complex preparation.
This medium supports the non-selective enrichment of Listeria monocytogenes, providing conditions for the bacteria to recover and multiply before being transferred to selective culture media, improving the detection and recovery of Listeria from test samples.

PRINCIPLES

BacterBag™ Dry Half Fraser Broth contains nutrients such as peptones, glucose, and yeast extract, providing nitrogen, carbon, and growth factors for bacteria. Additives like Tween 80, phosphate, and sodium help maintain stable pH and osmotic pressure, creating an ideal environment for bacterial recovery and multiplication before transferring to selective culture media.

Add the appropriate amount of sterile distilled water into the BacterBag™ Dry Half Fraser Broth packet according to the ratio provided on the packaging to create a complete solution..
Homogenize until the medium is completely dissolved.
Take 25 g of the sample to be analyzed and add it to a volumetric flask, then add 90 or 225 mL of the rehydrated BacterBag™ Dry Half Fraser Broth
Mix the contents thoroughly using an appropriate homogenizer.
Incubate the prepared culture at 35 – 37ºC for 18 – 24 hours.

RESULTS

This medium is used to recover and enrich bacteria in samples. After incubation, secondary culturing should be performed in other selective media to identify the bacteria.
