- PURPOSE OF USE
BacterBag™ Alkaline Saline Peptone Water (ASPW) is a media used as a primary and secondary enrichment medium to detect Vibrio spp. in human and animal food products as well as environmental samples.
Typical ingredients meet food microbiological standards NF EN ISO 21872-1.
- PRINCIPLE
BacterBag™ Alkaline Saline Peptone Water (ASPW) was first developed by Shread, Donovan and Lee3 as a non-selective liquid enrichment medium for the cultivation of Aeromonas species. Cruickshank reported that increased media pH could be used effectively in culturing Vibrio species that are tolerant to high alkalinity and chloride.
BacterBag™ Alkaline Saline Peptone Water (ASPW) complies with ISO/TS 21872-1:2007, which specifies a horizontal method for the detection of two major pathogenic Vibrio species that cause enteric disease in humans: V. parahaemolyticus and V. cholera. This method is applicable to products intended for human consumption and animal feed, as well as environmental samples in the food production and processing sector. Peptone provides carbon, nitrogen and essential nutrients to organisms. High sodium chloride concentrations, in addition to maintaining osmotic balance, also have the effect of inhibiting the accompanying microflora.
- Add Xg sample to 9XmL or 9Xg Alkaline Saline Peptone Water medium at ambient temperature.
- Incubate for about 6 ± 1 hour, at 41.5 ± 1°C for fresh samples or 37 ± 1°C for frozen or salted samples.
- Take 1mL of the incubated solution and inoculate it with 10ml of Alkaline Saline Peptone Water and incubate for about 18 ± 1 hour at 37 ± 1°C.
- From the obtained culture fluid, take 1 loop of culture rod and inoculate it onto the surface of TCBS Agar medium and a second selective isolation medium to obtain characteristic colonies.
- Incubate the inoculated plate at 37°C for about 24 ± 3 hours (or a suitable second selective medium).
- Identify characteristic colonies of pathogenic Vibrio species present on the plate. Select 5 types of colonies to culture on Saline Nutrient Agar medium for biochemical determination.
RESULT INTERPRETATION
- This medium is used for the recovery and enrichment of bacteria present in the sample. Thus, secondary culturing in other selective media is necessary for bacterial identification.
