INTENDED USE
- BacterBag™ Half Fraser Broth is used for the selective and differential enrichment (primary enrichment broth) of Listeria monocytogenes and Listeria spp. in food products, according to the NF EN ISO 11290-1 standard.
PRINCIPLES
- BacterBag™ Half Fraser Broth is used for the primary enrichment step.
- Enzymatic digest of animal tissues, enzymatic digest of casein, yeast extract, and meat extract provide essential nutrients for the growth of Listeria.
- The high sodium chloride content increases the selectivity of the medium.
- Phosphates act as buffers and maintain the medium’s pH.
- Esculin is hydrolyzed by Listeria into glucose and esculetin, the latter forming a black complex with iron ions provided by ferric ammonium citrate, which is added immediately before use. This also facilitates the growth of Listeria.
- Lithium chloride inhibits the growth of most intestinal cocci, which can also hydrolyze esculin.
- Nalidixic acid prevents the DNA replication of bacteria sensitive to this antimicrobial agent.
The growth of secondary gram-positive microorganisms is inhibited by acriflavine.
- Add 25g of the product to be analyzed into a bag containing BacterBag™ Half Fraser Broth.
- Mix thoroughly.
- Incubate for 18 – 24 hours at 35 – 37ºC.
- Note: BacterBag™ Half Fraser Broth can also be used as a diluent in the quantification of Listeria monocytogenes.
RESULTS
- This medium is used for the recovery and enrichment of bacteria present in the sample. Thus, secondary culturing in other selective media is necessary for bacterial identification.
