INTENDED USE

BacterStrain™ Eosin Eosin is primarily used in histology and cytology, particularly in the Hematoxylin and Eosin (H&E) staining method. It stains the cytoplasm, extracellular matrix, and other components of tissues, highlighting the structures of cells and tissues, and helping differentiate various components in tissue samples. Eosin is also used in other staining techniques to distinguish acidic components within cells.

PRINCIPLES

BacterStain™ Eosin is an acidic dye that binds to alkaline components in cells and tissues, especially to cytoplasm and basic proteins. In the H&E staining method, eosin stains these components red or pink, while hematoxylin stains the cell nuclei blue or purple. The combination of hematoxylin and eosin provides clear contrast, making cell and tissue structures more visible under a microscope.

Deparaffinize in 3 xylene baths, 5 minutes each.
Immerse through 4 alcohol baths: 100º – 95º – 80º – 70º, 15 dips in each bath.
Rinse with distilled water: 15 dips.
Stain the nucleus with Harris Hematoxylin: 3-5 minutes or longer.
Rinse under running water: 5-10 minutes.
Check the color of the nucleus under the microscope, if too dark, decolorize gently with alcohol-acid.
Rinse under running water: 1 minute.
Step 8: Stain with 1% Eosin: 1-2 minutes.
Rinse under running water: 1 minute.
Differentiation in 2 baths of 95º – 100º alcohol, 15 dips in each bath.
Immerse through 3 xylene baths, Bath I and II: 15 dips, Bath III: 5-10 minutes.
Mount the coverslip.

2	Smoothness	The specimen surface is continuous, even, not hollow or bare. Microscopic examination: Continuous microscopic fields without many empty microscopic fields, uniform brightness.
3	Thickness	Standard specimens have a thickness of about 0.04 mm. When the specimen is dry and not stained, it can be checked by placing a piece of paper with printed letters 4-5 cm below the specimen. If the letters are faint and readable, it is qualified. If the letters are illegible, it is thick. If the letters are too clear, it is thin.
