INTENDED USE
- BacterTube™ Urea Broth is recommended for the identification of bacteria based on urea utilization, particularly for differentiating Proteus, Salmonella, and Shigella species from clinical and non-clinical specimens.
PRINCIPLES
- This medium differentiates bacteria based on their ability to hydrolyze urea using the enzyme urease:
- Urea hydrolysis releases ammonia, increasing the medium’s pH, which changes the indicator color to pink/red.
- The high buffering capacity and minimal nutrients prevent false positives caused by protein hydrolysis.
- A negative control without urea ensures accurate results by ruling out false positive
- Prior to inoculation, allow medium to equilibrate to room temperature
- Use a sterile inoculation loop to transfer a sample from an isolated colony on the agar plate into the test tube containing the medium.
- Incubate the tube at 35°C.
- Examine the tube after 6 – 8 hours and again after 12 – 18 hours of incubation.
- Observe and record the results
RESULTS
- Positive Reaction: Pink to red color (urease activity detected).
- Negative Reaction: No color change (yellowish-orange remains)
