INTENDED USE
- BacterTube™ BHI Ge/Cl is a versatile nutrient medium recommended for cultivating and isolating various microorganisms, including bacteria, yeasts, and molds. The addition of antimicrobial agents creates a selective environment used for isolating pathogenic fungi from samples heavily contaminated with bacteria and saprophytic fungi. A range of Gram-positive and Gram-negative bacteria is inhibited by chloramphenicol, a broad-spectrum antimicrobial agent.
PRINCIPLES
- BacterTube™ BHI Ge/Cl with its main components including peptone, amino acids, nitrogen, sulfur, carbon, trace elements, and agar provides essential nutrients for bacterial growth. Dextrose serves as an energy source for the microorganism’s metabolic processes. Sodium chloride provides the necessary electrolytes. Combined with Chloramphenicol, a broad-spectrum antibiotic that inhibits a wide range of both Gram-positive and Gram-negative bacteria. Gentamicin, an aminoglycoside antibiotic, has bactericidal effects. It crosses the Gram-negative bacterial membrane through an active transport mechanism that is oxygen-dependent. This requirement for oxygen is the reason why aminoglycosides are not effective against anaerobic bacteria.
- Heat the tip of the inoculation loop until red in the flame, then gently heat the handle, and hold the loop vertically to ensure even heating.
- Inoculate the sample onto the surface of the slant agar by lightly placing the tip of the loop on the surface at the bottom of the tube.
- Then, streak the sample in a zig-zag pattern from the bottom of the test tube up to the top of the slant agar.
- Incubate under aerobic conditions at 35 – 37°C for 16 – 24 hours
RESULTS
- After incubating for the required time, typically 18-24 hours, observe the bacterial colonies growing on the surface of the medium.
- To identify the isolated bacteria, further appropriate tests must be conducted.
