INTENDED USE
- BacterTube™ Chocolate Agar (CAXV) is primarily used for preserving and transporting bacteria under warm environmental conditions. This medium helps maintain the viability of bacteria for a short period and ensures that bacteria requiring special nutrients can be preserved and transported without compromising sample quality. Using brown agar in slant tubes provides a larger surface area for bacterial growth and facilitates convenient sample transportation without affecting bacterial development..
PRINCIPLES
- Peptones included in the composition of the medium favor the excellent growth of colonies.
- Yeast extract is a source of vitamin B complex.
- Starch is a detoxifying agent and also an energy source.
- The heat treatment of blood in the medium lyses red blood cells, releasing growth factors such as hemin (X factor) and NAD (V factor), which are essential for the growth of certain bacteria.
- Heat the loop’s tip until it becomes red in the flame, then lightly heat the handle of the loop, ensuring the loop is uniformly hot.
- Inoculate the medium by gently placing the loop’s tip on the surface of the slant agar at the bottom of the tube.
- Next, streak in a zigzag pattern from the bottom of the tube to the top along the surface of the slant..
- Place the tube in the incubator under aerobic conditions at 35 – 37°C for 18 – 24 hours.
RESULTS
- After incubation for the required duration, observe the colonies growing on the surface of the plate for characteristic traits such as colony size, color, and morphology.
- Haemophilus influenzae: Smooth, greyish, translucent colonies.
- Neisseria gonorrhoeae: Small, shiny, grey colonies with a mucoid appearance.
- Identification of the isolated bacteria must be carried out through appropriate subsequent tests
