  • TraceMedia™ MacConkey Agar/Sabouraud Agar is a dual-compartment culture medium designed for the cultivation and isolation of Gram-negative bacteria and yeast from clinical specimens.

COMPOSITION
The medium consists of two compartments:

  • MacConkey Agar (MC): This compartment is used for the selective cultivation of Gram-negative bacteria, especially enteric bacteria such as those from the Pseudomonas genus. It inhibits the growth of Gram-positive bacteria and some difficult-to-grow Gram-negative bacteria due to the presence of crystal violet and bile salts in its composition. Bacteria that ferment lactose produce acid, leading to a decrease in the pH of the medium, which causes a color change to pink or red. Additionally, the acid produced from lactose fermentation reacts with bile salts to form precipitates, resulting in pink colonies with a surrounding bright zone. Bacteria that ferment lactose weakly still produce a pink or red color but do not form the bright zone. Non-lactose-fermenting bacteria retain their original color, and the colonies remain colorless.
  • Sabouraud Dextrose Agar (SDA): This compartment is used for the cultivation and isolation of yeast and mold. Its composition, containing peptone to provide nitrogen and vitamins required for organism growth, and its pH is adjusted to approximately 5.8 to enhance the growth of fungi and inhibit bacterial growth in clinical samples.

RESULTS:

  • After incubation for the specified time, typically 16-24 hours, observe bacterial colonies on the surface of the agar plate. READING RESULTS:

  • After incubation for the specified time, typically 16-24 hours, observe bacterial colonies on the surface of the agar plate.
  • Further identification of isolated bacteria must be conducted using appropriate tests.

The relevant samples are directly inoculated onto the agar plates.

  1. If the agar plates have been refrigerated, allow them to warm up to room temperature before inoculating the samples.
  2. Inoculate the samples onto the agar plates.
  3. Incubate the plates in an aerobic condition at 35 – 37°C for 16 – 24 hours.
