INTENDED USE
- BacterTest™ Kovac is a reagent used in the Indole Test reaction to determine the ability of bacteria to decompose tryptophan into indole, an organic compound that has a characteristic color when reacting with Kovac’s reagent.
PRINCIPLES
- BacterTest™ Kovac is based on the ability of certain bacteria to break down tryptophan into indole.
- Bacteria break down tryptophan into indole using the enzyme tryptophanase.
- Kovac’s reagent (p-Dimethylaminobenzaldehyde, HCl and isoamyl alcohol) reacts with indole to produce a red or pink color, which is an indication of a positive reaction (bacteria producing indole).
- Bacteria are cultured in a prepared medium containing tryptophan (usually Tryptone Broth) after 24 – 48 hours at 37°C.
- Adding 1 mL of Kovac’s reagent to the bacterial culture medium.
- Observing the color change of the medium, reading and recording the results.
RESULTS
- Observe the color change of the environment:
- Red = Positive (indole producing bacteria)
- Colorless or Yellow = Negative (bacteria do not produce indole).
