- INTENDED USE
BacterPlate™ Two-Compartment Medium BA/MC is used for the isolation and differentiation of bacteria, particularly Gram-negative bacteria, as well as bacteria capable of hemolysis or lactose fermentation. This medium is applied in microbiological research, clinical diagnostics, and microbiological quality control of water, food, pharmaceuticals, and animal-derived biological samples.
The packaging with semi-permeable Cellophane film helps balance the humidity of the environment during storage.
- PRINCIPLES
BA (Blood Agar) Compartment: This is a blood-based culture medium that helps differentiate bacteria based on their hemolytic properties. Hemolytic reactions can be categorized into three types: beta (β) hemolysis, alpha (α) hemolysis, and gamma (ɤ) hemolysis. These hemolytic reactions help identify the characteristics of bacteria such as Streptococcus and Listeria.
MC (MacConkey Agar) Compartment: MacConkey Agar is a selective medium used to isolate Gram-negative bacteria, particularly enteric bacteria. Bacteria capable of lactose fermentation will form red or pink colonies, while non-lactose fermenters will form colorless or transparent colonies
INSTRUCTIONS FOR USE
- Allow the agar plates to reach room temperature before use.
- Dry the agar plates in an incubator with the lid slightly open.
- Inoculate the sample by spreading it evenly on the surface of the agar using a sterile inoculating loop.
- Incubate at 35–37°C for 18–24 hours.
RESULTS
- BA Compartment:
- Beta Hemolysis (β): Colonies will appear with a complete hemolytic zone (clear zone) around them. Beta-hemolytic bacteria include Streptococcus pyogenes and Listeria.
- Alpha Hemolysis (α): Colonies will have a narrow, greenish or faint hemolytic zone. Alpha-hemolytic bacteria include Streptococcus pneumoniae.
- Gamma Hemolysis (ɤ): No hemolytic zone, e.g., Enterococcus faecalis.
- MC Compartment:
- Lactose Positive: Lactose-fermenting bacteria will form red colonies, surrounded by a precipitate of bile salts.
- Lactose Negative: Non-lactose fermenting bacteria will form colorless or transparent colonies.
