INTENDED USE

BacterPlate™ Two-Compartment Medium TCBS/SS is used for the isolation and differentiation of enteric pathogens such as Vibrio and Salmonella, particularly in fecal, seafood, and animal-derived samples. This medium helps differentiate Vibrio cholerae from other Vibrio species and simultaneously isolates Salmonella and Shigella.
The packaging with semi-permeable Cellophane film helps balance the humidity of the environment during storage.

PRINCIPLES

BacterPlate™ Two-Compartment Medium TCBS/SS:

TCBS (Thiosulfate Citrate Bile Sucrose Agar) Compartment: This medium is used for the isolation and differentiation of Vibrio species, particularly Vibrio cholerae. The medium contains components that inhibit the growth of Gram-positive bacteria and some Gram-negative bacteria. The fermentation of sucrose by Vibrio produces acid, causing the medium to turn yellow.
SS (Salmonella Shigella Agar) Compartment: This medium supports the growth of Salmonella and Shigella while inhibiting the growth of other bacteria such as coliforms. Bacteria that produce hydrogen sulfide (H₂S) form colonies with black centers, while non-lactose fermenters form colorless colonies.

Allow the agar plates to reach room temperature before use.
Dry the agar plates in an incubator with the lid slightly open.
Inoculate the sample by spreading it evenly on the surface of the agar using a sterile inoculating loop.
Incubate at 35 – 37°C for 18 – 24 hours.

RESULTS

TCBS Compartment:

Vibrio cholerae: Yellow, flat colonies, 2–3 mm in diameter.
Other Vibrio species (Vibrio fluvialis, furnissii, V. vulnificus, V. parahaemolyticus, V. mimicus): Green, flat colonies.
Pseudomonas, Aeromonas, Enterobacteriaceae: Blue or colorless colonies.

SS Compartment:
