INTENDED USE
- BacterTube™ Malonate Broth h is recommended for use in the differentiation of Enterobacteriaceae on the basis of malonate utilization.
PRINCIPLES
- The medium distinguishes malonate-positive organisms (e.g., Klebsiella pneumoniae) from malonate-negative organisms (e.g., Escherichia coli).
- Malonate and Ammonium Sulfate: Organisms that utilize both produce sodium hydroxide, causing alkalinity and a color change to light blue or Prussian blue.
- Bromothymol Blue: pH indicator; turns blue in alkaline conditions and yellow in acidic environments due to glucose fermentation.
- Yeast Extract and Dextrose: Provide minimal growth for organisms unable to utilize malonate
- Prior to inoculation, allow medium to equilibrate to room temperature
- Using a light inoculum from an 18 – 24 hour pure culture, inoculate the tube.
- Incubate the tube with loosened caps at 35ºC in an aerobic atmosphere for 24 – 48 hours
- Observe for alkalization (blue color) at 24 and 48 hours
RESULTS
- Positive Test: Blue color indicates malonate utilization.
- Negative Test: Green or yellow color (due to glucose fermentation)
