INTENDED USE
- BacterTube™ Phenylalanine Deaminase Agar (PAD) is recommended for differentiating Proteus and Providencia species from other members of the Enterobacteriaceae family based on their ability to deaminate phenylalanine to form phenylpyruvic acid.
PRINCIPLES
- Proteus and Providencia species can deaminate phenylalanine to produce phenylpyruvic acid.
- Meat extract provides essential growth nutrients.
- Sodium chloride maintains osmotic balance.
- After incubation, phenylpyruvic acid reacts with ferric chloride to produce a green color, indicating a positive result
- Inoculate the BacterTube™ Phenylalanine Deaminase Agar (PAD) with the cultured bacteria.
- Incubate the inoculated tubes at 35 – 37°C for 24 hours.
- After incubation, add the reagent to the tubes.
RESULTS
- Positive result: The medium turns dark green. This color change is due to degradation products produced by phenylalanine. These products react with the iron compounds in the acidic environment, resulting in the dark green color.
- Negative result: The color of the medium remains unchanged, staying straw yellow (no PPA to react with FeCl3)
