- The heated blood causes the medium to turn brown and release factors X (Hemin) and V (Nicotinamide adenine dinucleotide, NAD) essential for the growth of Neisseria gonorrhoeae, Neisseria meningitidis, and Haemophilus spp.
- After incubation for the required time, observe the growth of colonies on the surface of the plate and their characteristics.
