INTENDED USE
- BacterTube™ Alkaline Peptone is used as a primary and secondary enrichment medium for the detection of Vibrio spp., including Vibrio parahaemolyticus, Vibrio cholerae, and Vibrio vulnificus, from food products (human and animal) and environmental samples. The medium complies with the food microbiology standard NF EN ISO 21872-1.
PRINCIPLES
- BacterTube™ Alkaline Peptone creates a high pH (8,6 ± 0,2) and saline environment that favors the growth of Vibrio spp. while inhibiting competing bacteria. Peptone provides essential nutrients, and sodium chloride mimics marine conditions, enhancing the recovery of halophilic Vibrio spp.. The two-step enrichment process increases the concentration of target organisms for easier detection in food and environmental samples.
Detection of Vibrio parahaemolyticus and Vibrio cholerae
- Aseptically add X g of the sample to 9X mL of BacterTube™ Alkaline Peptone, mix thoroughly.
- Incubate for 6 ± 1 hours at:
- 37 ± 1°C for deep-frozen, dried, or salted products.
- 5 ± 1.0°C for fresh products.
- Transfer 1 mL of the culture into 10 mL of fresh BacterTube™ Alkaline Peptone.
- Incubate for 18 ± 1 hours at:
- 5 ± 1.0°C for Vibrio parahaemolyticus and Vibrio cholerae enrichment.
- 37 ± 1°C for Vibrio vulnificus
Note: For general Vibrio spp. detection, all enrichments should be incubated at 37°C.
