INTENDED USE
- BacterPlate™ Chocolate Agar with Bacitracin (CAHI) is a selective, enriched culture medium designed for the isolation of Haemophilus influenzae and other fastidious bacteria from clinical specimens. The addition of Bacitracin suppresses the growth of contaminating flora, enhancing the recovery of H. influenzae. This medium is widely used in clinical microbiology and diagnostic laboratories for targeted pathogen identification.
- The typical composition of base medium (Columbia Agar) corresponds to that defined in European (EP), United States (UP) and Japanese (JP) Pharmacopeias.
- The packaging with semi-permeable Cellophane film helps balance the humidity of the environment during storage.
PRINCIPLE
- Special peptone provides essential nutrients, and sodium chloride acts as a pH buffer while starch neutralizes toxic fatty acids that may be present in the agar.
- Isovitalex is a supplement providing factor V (nicotinamide adenine dinucleotide, NAD) for Haemophilus species.
- Bacitracin is a polypeptide antibiotic that inhibits gram-positive bacteria, Neisseria, and Haemophilus spp.
- Allow the agar plates to reach room temperature. Dry the plates in an incubator by slightly opening the lid.
- Inoculate in order to obtain isolated colonies.
- Incubate at 37 °C, 5 – 10% CO2 for 18 – 24 hours in optimal conditions for the culture of the inoculated germs.
RESULTS
- After incubation for the required duration, observe the colonies growing on the surface of the plate for characteristic traits such as colony size, color, and morphology.
- Haemophilus influenzae: Small, greyish, smooth colonies with a moist appearance.
- Other: Growth of other bacteria should be suppressed due to the presence of Bacitracin.
- Identification of the isolated bacteria must be carried out through appropriate subsequent tests
